The Food and Drug Administration approved the anti-cancer drug crizotinib and it is for the m-
etastatic non-small cell lung cancer (NSCLC) patients. Their tumors are very anaplastic lymp-
homa (ALK)-positive by a FDA test. Their rates were high regarding the progressed free survi-
al (PFS) rate and overall response rate (ORR).
The treatment helped patients tremendously. Conductors did a trial that was randomized, activ-
e-controlled, open-labeled, & also multinational. It enrolled 347 patients with ALK-positive, met-
astatic non-small cell lung cancer. Those ill people had already progressed following platinum-b-
ased chemotherapy.
Some of them were chosen randomly to consume 250 mg of crizotinib two times a day. The oth-
er participants got the chemotherapy pemetrexed or docetaxel. Patients who were given crizotin-
ib showed significantly prolonged PFS against the folks who were given the chemotherapy. The
median PFS was 7.7 months after crizotinib consumption and 3 months for the earthlings who g-
ot chemotherapy.
The overall response rate was 65% for the patients who took crizotinib and 20% for the chemot-
herapy-receivers. Since that was the case, the Food and Drug Administration handed the accel-
erated approval for crizotinib (Xalkori).
The anti-cancer drug afatinib (Gilotrif) got the seal of approval from the FDA too. It's for the people
with metastatic non-small cell lung cancer (NSCLC). Their tumors have the epidermal growth fact-
or receptors (EGFR) exon 19 deletions or exon 22 (L8558R) substitutions-mutations. The Food &
Drug Administration concurrently gave their approval for the therascreen EGFR RGQ PCR kit that
locates the molecular markers.
The FDA got the results from a trial that was controlled and randomized with Lux-Lung 3. The res-
earchers chose 345 participants with metastatic NSCLC and their tumors had EGFR mutations. S-
ome of the ill folks took 40 g of afatinib once a day through their mouths. The other folks were giv-
en the treatment pemetrexed and cisplatin.
The conductors made sure to stratify the randomization according to status of each epidermal gro-
wth factor receptor-mutation. The categories were Asian and non-Asian. The researchers only put
the racial part in to test the treatments. The researchers are not hateful at all. The median progres-
sion free survival (PFS) was 11.1 months for the afatinib-takers and 6.9 months for the chemothe-
rapy-receivers.
The objective response rate was 50.4% for the patients who given the afatinib and 19.1% for the p-
eople who had the chemotherapy. There was no major difference in overall survival. Some of the fo-
lks with exon 19 deletions and L8558R mutations (n=308) median PFS was 13.6 months, after tak-
ing afatinib.
The patients' PFS was 6.9% after receiving chemotherapy. A lot of cancer patients had such mutat-
ions, but 11% had other types of mutations. Gilotrif (afatinib) is a drug to handle EGFR and the effe-
cts of it can not be reversed. It is proven to manage activity versus mutations that have become resi-
stant to two reversible TKIs erlotinib & gefitinib. Thank God for Gilotrif because it's really helping the
cancer patients thoroughly.
